Mouse Interleukin 2 ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of mouse Interleukin-2 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. An antibody specific for IL-2 is pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added, IL-2 is captured by the immobilised antibody, and bound analyte is detected with a biotinylated anti-IL-2 antibody followed by HRP-avidin conjugate. TMB substrate is used to visualise the HRP reaction, producing a blue-coloured product that turns yellow after the acidic stop solution is added; absorbance is read at 450 nm on a colorimetric microplate reader and IL-2 concentration is calculated from the standard curve.

The assay has a stated product range of 15.6-1000 pg/ml and a sensitivity of <6.0 pg/ml, with a total assay time of 4h 30m. Spike-and-recovery data (n=5 per matrix) are reported as: serum 80%-102% (average 91%), EDTA plasma 81%-100% (average 90%), and heparin plasma 80%-89% (average 84%). Dilutional linearity values for serum, EDTA plasma and heparin plasma (n=5 each) are provided at 1:2, 1:4, 1:8 and 1:16 dilutions.

Kit components comprise a pre-coated 96-well microplate (12 x 8 well strips), lyophilised standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and plate sealers (5 adhesive strips). Components are stored at +4°C or -20°C according to the instructions; the kit should not be used past its expiration date.
