Mouse Interferon Regulatory Factor 1 ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) enables the in vitro quantitative determination of mouse Interferon Regulatory Factor 1 (IRF1) in tissue homogenates, cell lysates, or other biological fluids. The assay uses a pre-coated 96-well microplate configured as 12 x 8 well strips, with absorbance read on a colorimetric microplate reader at 450 nm.

Standards and test samples are added to the wells, where IRF1 is captured by an immobilized antibody. After washing, a biotinylated anti-IRF1 antibody is added, followed by an HRP-avidin conjugate. TMB substrate produces a blue product that turns yellow upon addition of acidic stop solution; the absorbance is proportional to the amount of IRF1 captured. Reported sensitivity is <0.058 ng/ml with an assay range of 0.156-10 ng/ml. Total assay time is 4 hours 30 minutes, and components are stored at +4°C or -20°C as instructed. The kit should not be used past its expiration date.

Manufacturer validation data report mean recovery of 91% from serum (range 80%-102%, n=5), 90% from EDTA plasma (range 81%-100%, n=5), and 84% from heparin plasma (range 80%-89%, n=5). Dilution linearity tested at 1:2, 1:4, 1:8 and 1:16 dilutions gave the following ranges (all n=5): serum 87-91%, 87-107%, 74-101% and 92-97%; EDTA plasma 90-105%, 84-101%, 90-101% and 79-108%; heparin plasma 84-95%, 92-105%, 82-105% and 89-91%.
