Description
Antibodies.com supplies a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) intended for the in vitro quantitative determination of mouse Insulin Receptor Substrate 1 (IRS1) in tissue homogenates, cell lysates, or other biological fluids. The assay uses the sandwich enzyme immunoassay technique. An antibody specific for IRS1 is pre-coated onto a 96-well microplate; standards and samples are added and any IRS1 present is captured by the immobilized antibody. After washing, a Biotinylated Anti-IRS1 Antibody binds the captured analyte, followed by an HRP-Avidin conjugate. TMB substrate is then added, and the resulting blue product changes to yellow upon addition of an acidic stop solution. The density of yellow is proportional to the amount of IRS1 captured, and concentration is calculated by reading absorbance at 450 nm against a standard curve.
Key specifications include an assay range of 0.312–20 ng/ml and a sensitivity of <0.124 ng/ml, with a total assay time of 4 hours 30 minutes. The platform is a pre-coated microplate arranged as 12 × 8-well strips, read on a colorimetric microplate reader.
The product is reactive with mouse and is suited to cell lysates and tissue homogenates. Kit components are stored at +4 °C or −20 °C as instructed, and the kit should not be used past its expiration date.
