Mouse Insulin Promoter Factor 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of mouse PDX1 in tissue homogenates, cell lysates, or other biological fluids. The assay uses a pre-coated 96-well microplate read on a colorimetric microplate reader at 450 nm.

An antibody specific for PDX1 is pre-coated onto the plate. Samples and standards are added and PDX1 is captured by the immobilized antibody. After washing, a Biotinylated Anti-PDX1 Antibody binds the captured target, followed by an HRP-Avidin conjugate. TMB substrate produces a blue colour that turns yellow upon addition of stop solution; the absorbance at 450 nm is proportional to PDX1 concentration and is quantified against a standard curve.

The reported assay range is 0.156–10 ng/ml with a sensitivity of <0.053 ng/ml. Total assay time is 4 hours 30 minutes. Recovery has been validated across five samples per matrix: serum 80%–102% (average 91%), EDTA plasma 81%–100% (average 90%), and heparin plasma 80%–89% (average 84%). Dilution linearity was tested at 1:2, 1:4, 1:8, and 1:16 across the same sample types.
