This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit from Antibodies.com enables the in vitro quantitative determination of mouse Insulin Like Protein 6 (INSL6) in serum, plasma, tissue homogenates, and other biological fluids. The assay uses a pre-coated 96-well microplate to which an INSL6-specific antibody has been immobilised. Standards and test samples are pipetted into the wells and any INSL6 present is captured by the immobilised antibody. After washing, a biotinylated anti-INSL6 detection antibody is added, followed by an HRP-avidin conjugate. TMB substrate solution is then introduced, producing a blue colour that turns yellow on addition of the acidic stop solution. The intensity of the yellow colour is proportional to the amount of INSL6 captured, and concentration is calculated from the optical density read at 450 nm on a colorimetric microplate reader.

According to the manufacturer, sensitivity is <30 pg/ml and the assay range is 78.1–5000 pg/ml. In spike-and-recovery testing with n=5 per matrix, average recovery was 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%), and 84% for heparin plasma (range 80–89%). Linearity-of-dilution data are provided across 1:2, 1:4, 1:8 and 1:16 dilutions for all three sample types. The total assay time is approximately 4 hours 30 minutes. Kit components include the pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer concentrate, sample dilution buffer, TMB substrate, stop solution and plate sealers, which should be stored at +4 °C or −20 °C as instructed. The product is intended for research use only.
