The assay uses a 96-well microtiter plate pre-coated with an antibody specific for IGFBP6; standards and samples are added to the wells and the target is captured by the immobilized antibody.

After washing, a biotinylated anti-IGFBP6 detection antibody is added, followed by an HRP-avidin conjugate and TMB substrate. The enzymatic reaction produces a blue product that turns yellow upon addition of acidic stop solution; absorbance is read at 450 nm in a colorimetric microplate reader and the IGFBP6 concentration is calculated against the standard curve. The stated product range is 6.25–400 ng/ml with a sensitivity of 2.7 ng/ml, and the total assay time is 4 hours 30 minutes.
