Mouse Insulin Like Growth Factor Binding Protein 4 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of mouse Insulin Like Growth Factor Binding Protein 4 (IGFBP4) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with a total assay time of 4h 30m, and is read on a colorimetric microplate reader at 450 nm. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.

In the procedure, standards and samples are added to wells pre-coated with an antibody specific for IGFBP4. Following incubation and washing, a biotinylated anti-IGFBP4 antibody binds the captured IGFBP4, then HRP-Avidin conjugate is added. TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow after addition of acidic stop solution. The intensity of the yellow colour is proportional to the amount of IGFBP4 captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm and referring to the standard curve.

The standard product range is 15.6-1000 pg/ml with a sensitivity of 6.4 pg/ml. Manufacturer-reported recovery averages include 91% in serum (range 80%-102%, n=5), 91% in EDTA plasma (range 81%-101%, n=5), and 84% in heparin plasma (range 80%-89%, n=5). Linearity upon dilution across 1:2, 1:4, 1:8 and 1:16 is reported within ranges that vary by sample matrix.
