From Antibodies.com, this sandwich enzyme-linked immunosorbent assay (sELISA) is designed for the in vitro quantitative determination of mouse IKB alpha in tissue homogenates, cell lysates, or other biological fluids. The Mouse IKB alpha ELISA Kit employs an antibody specific for IKB alpha pre-coated onto a microplate. Standards and test samples are added to the wells, followed by incubation with a Biotinylated Anti-IKB alpha Antibody and an HRP-Avidin conjugate. TMB substrate is then added; the HRP enzymatic reaction produces a blue product that turns yellow upon addition of the stop solution, and the absorbance is measured at 450 nm using a colorimetric microplate reader.

The quantitative assay has a reported range of 0.312–20 ng/ml and a sensitivity of <0.113 ng/ml, with a total assay time of 4 hours 30 minutes. The supplied kit includes a pre-coated microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers. Recovery data indicate average values of 91% for serum (range 80%–102%), 90% for EDTA plasma (range 81%–100%), and 84% for heparin plasma (range 80%–89%) (n=5), with linearity data across dilutions of 1:2, 1:4, 1:8, and 1:16 in each matrix. Kit components should be stored at +4°C or −20°C as instructed, and the kit must not be used past its expiration date.
