Mouse Indoleamine-2,3-Dioxygenase 2 ELISA Kit

This sandwich ELISA from Antibodies.com enables the in vitro quantitative determination of mouse Indoleamine-2,3-Dioxygenase 2 (IDO-2) in serum, plasma, tissue homogenates, and other biological fluids. The assay uses the sandwich enzyme immunoassay technique, in which an antibody specific for IDO-2 is pre-coated onto a 96-well microplate. Standards and samples are added to the wells, followed by a Biotinylated Anti-IDO-2 Antibody, an HRP-Avidin conjugate, and a TMB substrate solution; the resulting colour change is read as absorbance at 450 nm on a microplate reader, with IDO-2 concentration calculated against a standard curve.

The quantitative assay has a reported detection range of 0.781–50 ng/ml and a sensitivity of <0.32 ng/ml. It runs on a pre-coated microplate platform of 12 × 8 well strips, with a total assay time of 4 h 30 min, and detection is colorimetric. Kit components are stored at +4 °C or −20 °C as instructed, and the kit should not be used past its expiration date.

Validation data are provided for serum, EDTA plasma, and heparin plasma matrices. Reported recovery ranges are 80%–102% for serum (n=5), 81%–100% for EDTA plasma (n=5), and 80%–89% for heparin plasma (n=5). Dilution linearity has been evaluated at 1:2, 1:4, 1:8, and 1:16 dilutions, with serum recoveries of 87%–91%, 87%–107%, 74%–101%, and 92%–97% (n=5); EDTA plasma recoveries of 90%–105%, 84%–101%, 90%–101%, and 79%–108% (n=5); and heparin plasma recoveries of 84%–95%, 92%–105%, 82%–105%, and 89%–91% (n=5).
