Description
From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) is intended for the in vitro quantitative determination of mouse Hypoxia Inducible Factor 2 Alpha (HIF-2 alpha) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.
The assay uses a microplate pre-coated with an antibody specific for HIF-2 alpha. Standards and samples are added to the wells and HIF-2 alpha is captured by the immobilised antibody, then detected using a Biotinylated Anti-HIF-2 alpha Antibody, HRP-Avidin conjugate and TMB substrate. The enzymatic reaction produces a yellow product after addition of acidic stop solution, with absorbance read at 450 nm on a colorimetric microplate reader. The standard curve spans 0.156–10 ng/ml with a reported sensitivity of 0.053 ng/ml, and the assay is completed in 4 hours 30 minutes.
In spike-and-recovery testing (n=5), serum samples gave an average of 91% (range 83–100%), EDTA plasma 96% (range 90–103%) and heparin plasma 94% (range 90–99%). Dilution linearity was demonstrated at 1:2, 1:4, 1:8 and 1:16 in matched serum, EDTA plasma and heparin plasma matrices. Kit components are stored at +4°C or −20°C as instructed and should not be used past the stated expiration date.
