Antibodies.com supplies a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) intended for the in vitro quantitative determination of mouse Heparanase 1 in serum, plasma or other biological fluids. The kit is configured for use with a colorimetric microplate reader and uses a pre-coated microplate into which standards and test samples are added; an antibody specific for Heparanase 1 is immobilised on the plate to capture the analyte. Following incubation and washing, a Biotinylated Anti-Heparanase 1 Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue colour that turns yellow on addition of acidic stop solution. The density of the yellow product is proportional to the amount of Heparanase 1 captured, and concentration is calculated from the O.D. absorbance read at 450 nm against a standard curve. Reported sensitivity is <6.5 pg/ml with an assay range of 15.6–1000 pg/ml, and the total assay time is 4h 30m.

Storage guidance directs users to keep kit components at +4°C or −20°C as instructed and not to use the kit past its expiration date. Reported recovery values are 80%–102% (average 91%, n=5) in serum, 81%–100% (average 90%, n=5) in EDTA plasma, and 80%–89% (average 84%, n=5) in heparin plasma. Linearity data are reported across 1:2, 1:4, 1:8 and 1:16 dilutions for each of these sample types.
