From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of mouse Heat Shock Protein 90kDa Alpha A1 (HSP90 alpha) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for HSP90 alpha that has been pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, followed by incubation with a Biotinylated Anti-HSP90 alpha Antibody and then an HRP-Avidin conjugate. After further incubation and washing, TMB substrate is added to produce a blue colour that changes to yellow upon addition of stop solution; the intensity of yellow is proportional to the amount of HSP90 alpha captured in each well.

Quantitative measurement is performed by reading the O.D. absorbance at 450nm in a colorimetric microplate reader and referring to the standard curve. The kit has a product range of 78.1-5000 pg/ml and a sensitivity of <31 pg/ml. Total assay time is 4h 30m, and kit components should be stored at +4°C or -20°C as instructed; the kit should not be used past its expiration date.

Recovery performance was assessed across sample matrices. Average recoveries were 91% in serum (range 80%-102%, n=5), 91% in EDTA plasma (range 81%-101%, n=5), and 84% in heparin plasma (range 80%-89%, n=5). Dilution linearity was demonstrated at 1:2, 1:4, 1:8, and 1:16 dilutions across serum (74-107%), EDTA plasma (79-108%), and heparin plasma (82-105%) sample types (n=5).
