From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) is intended for the in vitro quantitative determination of mouse Heat Shock Protein 40 (HSP40), also referred to in the literature as DNAJB1, in tissue homogenates, cell lysates, and other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added to the wells and HSP40 is captured by an immobilised antibody specific for the target, then detected using a biotinylated anti-HSP40 antibody, an HRP-avidin conjugate, and TMB substrate. The enzymatic reaction produces a colour change read at 450 nm in a microplate reader, with concentration calculated against the standard curve. Total assay time is 4 hours 30 minutes.

The kit reports a sensitivity of <0.215 ng/ml and an assay range of 0.625-40 ng/ml. Validation data show average recovery values of 91% for serum (n=5, range 83%-100%), 94% for EDTA plasma (n=5, range 91%-98%), and 94% for heparin plasma (n=5, range 90%-99%). Dilution linearity is reported at 1:2, 1:4, 1:8, and 1:16 across serum, EDTA plasma, and heparin plasma samples.

Kit components include a pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers. Components are stored at +4°C or -20°C as instructed in the protocol manual, and the kit should not be used past its expiration date.
