Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit is intended for the in vitro quantitative determination of mouse Heat Shock 60kD Protein 1 (HSP60, Chaperonin) in serum, plasma, tissue homogenates, cell lysates, cell culture supernatants and other biological fluids. The assay employs a pre-coated 96-well microplate (12 x 8 well strips) onto which an antibody specific for HSP60 has been immobilised; standards and test samples are added, followed by a Biotinylated Anti-HSP60 Antibody, HRP-Avidin conjugate and TMB substrate, with the coloured product read at 450 nm on a colorimetric microplate reader.

Quantitative performance characteristics include a stated measuring range of 0.234-15 ng/ml and a sensitivity of <0.11 ng/ml, with a total assay time of 4h 30m. Reported recovery in spiked mouse serum (n=5) averages 91% (range 80-102%), in EDTA plasma averages 90% (range 81-100%), and in heparin plasma averages 84% (range 80-89%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions (n=5 each) is reported as 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma.

Supplied components include the Pre-Coated 96 Well Microplate (12 x 8 well strips, -20°C), lyophilised standard (2 vials, -20°C), Detection Solution A (120 µl, -20°C), Detection Solution B (120 µl, -20°C), 30X Wash Buffer (20 ml, +4°C), Sample Dilution Buffer (45 ml, -20°C), TMB Substrate (9 ml, +4°C), Stop Solution (6 ml, +4°C) and 5 adhesive plate sealers. Components are stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
