Mouse Haptoglobin ELISA Kit

This sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of mouse Haptoglobin in serum, plasma or other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for Haptoglobin. Standards and samples are added to the wells, where Haptoglobin is captured by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-Haptoglobin Antibody is added, then HRP-Avidin conjugate and TMB substrate. The enzymatic reaction produces a blue colour that changes to yellow upon addition of acidic stop solution, with absorbance read at 450 nm in a colorimetric microplate reader.

The assay has a range of 31.2–2000 ng/ml and a sensitivity of <13.9 ng/ml, with a total assay time of 4 hours 30 minutes on a pre-coated microplate of 12 x 8 well strips. Recovery in serum (n=5) is 89–101% with an average of 95%; in EDTA plasma (n=5) 91–101% with an average of 96%; and in heparin plasma (n=5) 91–101% with an average of 96%. Dilutional linearity has been demonstrated at 1:2, 1:4, 1:8 and 1:16 dilutions in serum, EDTA plasma and heparin plasma (n=5 per matrix).

Kit components include a pre-coated 96-well microplate (12 x 8 well strips), lyophilized standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and 5 adhesive plate sealers. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
