From Antibodies.com, this Mouse Growth Hormone Releasing Hormone (GHRH) ELISA Kit is a sandwich enzyme-linked immunosorbent assay designed for the in vitro quantitative determination of mouse GHRH in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids.

The assay uses a 96-well microplate pre-coated with a capture antibody specific for GHRH. Standards and test samples are added to the wells, where GHRH is bound by the immobilized antibody. After washing, a Biotinylated Anti-GHRH Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate solution is then added; the resulting enzymatic reaction produces a blue product that changes to yellow upon addition of acidic stop solution. The intensity of yellow, read at 450 nm in a microplate reader, is proportional to the amount of GHRH captured, and the concentration is calculated by reference to a standard curve.

The assay has a working range of 15.6-1000 pg/ml and a sensitivity of <4.34 pg/ml. Reported average recoveries are 91% for serum (range 80-102%, n=5), 90% for EDTA plasma (range 81-100%, n=5), and 84% for heparin plasma (range 80-89%, n=5). The assay is performed on a pre-coated microplate platform with 12 x 8 well strips and takes approximately 4 hours 30 minutes to complete. Detection is colorimetric, read on a standard microplate reader, and kit components should be stored at +4°C or -20°C as instructed. The kit should not be used past its expiration date.
