Mouse Glypican 1 ELISA Kit

Designed for the in vitro quantitative determination of mouse Glypican 1/GPC1, this sandwich ELISA from Antibodies.com is intended for use with serum, plasma, tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for Glypican 1/GPC1 pre-coated onto a 96-well microplate. Standards and samples are added to the wells, followed by incubation with a Biotinylated Anti-Glypican 1/GPC1 Antibody and then an HRP-Avidin conjugate. TMB substrate is used to visualise the enzymatic reaction, producing a blue product that changes to yellow upon addition of stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader, and the concentration of Glypican 1/GPC1 is calculated by reference to the standard curve.

Sensitivity is reported at <35 pg/ml with an assay range of 78.1-5000 pg/ml. Total assay time is 4h 30m and the platform is a pre-coated microplate arranged as 12 x 8 well strips.

Spike-and-recovery (average) values were 91% for serum (range 80%-102%, n=5), 90% for EDTA plasma (range 81%-100%, n=5) and 84% for heparin plasma (range 80%-89%, n=5). Dilution linearity across 1:2 to 1:16 dilutions gave ranges of 87-91% to 92-97% for serum, 90-105% to 79-108% for EDTA plasma, and 84-95% to 89-91% for heparin plasma (n=5 each). Kit components include the pre-coated plate, lyophilized standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, with storage between +4°C and -20°C as specified per component.
