Mouse Glutaminase 2 ELISA Kit

A sandwich ELISA from Antibodies.com enables the in vitro quantitative determination of mouse GLS2 (Glutaminase 2) in serum, plasma, tissue homogenates or other biological fluids. The kit employs a microtiter plate pre-coated with an antibody specific for GLS2. Standards and test samples are added to the wells, where GLS2 is captured by the immobilised antibody. After washing, a biotinylated anti-GLS2 antibody is added, followed by an HRP-avidin conjugate.

The HRP enzymatic reaction is visualised using TMB substrate, producing a blue-coloured product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm using a colorimetric microplate reader, and the concentration of GLS2 is calculated by reference to a standard curve. The assay has a sensitivity of <0.25 ng/ml and a measurement range of 0.625-40 ng/ml, with a total assay time of 4h 30m. Kit components should be stored at +4°C or -20°C as instructed and not used past the expiration date.

Spike-and-recovery testing in five samples per matrix gave averages of 91% in serum (range 80%-102%), 90% in EDTA plasma (range 81%-100%) and 84% in heparin plasma (range 80%-89%). Dilution linearity was demonstrated at 1:2, 1:4, 1:8 and 1:16 in serum, EDTA plasma and heparin plasma.
