A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com enables the in vitro quantitative measurement of mouse Glucose 6 Phosphate Dehydrogenase in serum, plasma, or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) immobilised with an antibody specific to the target. Standards and test samples are added to the wells, where the analyte is captured by the immobilised antibody. After washing, a Biotinylated Anti-Glucose 6 Phosphate Dehydrogenase Antibody binds the captured analyte, followed by incubation with an HRP-Avidin conjugate. TMB substrate solution is added to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of target captured, and concentration is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader and referring to a standard curve.

The quantitative assay has a working range of 0.312-20 ng/ml and a sensitivity of 0.137 ng/ml, with a total assay time of 4h 30m. Kit components are stored at +4°C or -20°C as instructed and should not be used after the expiration date. Spiking recovery has been demonstrated in serum (average 91%, range 80-102%, n=5), EDTA plasma (average 90%, range 81-100%, n=5), and heparin plasma (average 84%, range 80-89%, n=5). Dilution linearity has been demonstrated at 1:2, 1:4, 1:8, and 1:16 dilutions across serum, EDTA plasma, and heparin plasma (n=5).
