Mouse Fibulin 2 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) supplied by Antibodies.com, intended for the in vitro quantitative determination of mouse Fibulin 2 in serum, plasma and other biological fluids. The assay uses an antibody specific for Fibulin 2 pre-coated onto a 96-well microtiter plate. Standards and samples are added to the wells; captured Fibulin 2 is bound by a Biotinylated Anti-Fibulin 2 Antibody, and an HRP-Avidin conjugate is then introduced. A TMB substrate yields a blue colour that converts to yellow upon addition of acidic stop solution; absorbance is read at 450 nm with a microplate reader.

The assay covers 46.8-3000 pg/ml with a sensitivity of <21.45 pg/ml and a total run time of 4 hours 30 minutes. The platform is a pre-coated 96-well microplate arranged as 12 x 8 well strips, with colourimetric detection. Supplied components include the pre-coated microplate, lyophilized standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and adhesive plate sealers. Kit components are stored at +4 °C or -20 °C as instructed, and the kit should not be used past its expiration date.

Sample recovery was assessed in serum (n=5, average 91%, range 80-102%), EDTA plasma (n=5, average 90%, range 81-100%) and heparin plasma (n=5, average 84%, range 80-89%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions was 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma.
