Mouse Fibroblast Growth Factor 6 ELISA Kit

This sandwich enzyme-linked immunosorbent assay (sELISA) from Antibodies.com is intended for the in vitro quantitative determination of mouse Fibroblast Growth Factor 6 (FGF6) in serum, plasma and other biological fluids. The kit uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with an antibody specific for FGF6 immobilised on the plate surface.

Standards and test samples are added to the wells; FGF6 present in each sample is captured by the immobilised antibody. After washing, a biotinylated anti-FGF6 antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then introduced, and the HRP enzymatic reaction is visualised as a blue colour that changes to yellow upon addition of the acidic stop solution. The intensity of the yellow colour is proportional to the amount of FGF6 captured in each well. Absorbance is read at 450 nm in a colorimetric microplate reader, and the FGF6 concentration is calculated by reference to a standard curve. The assay range is 15.6-1000 pg/ml with a sensitivity of <7.4 pg/ml, and the total assay time is 4h 30m.

Kit components include a pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, to be stored at +4°C or -20°C as instructed. The kit should not be used past its expiration date. Reported recovery values are: serum (n=5) 80-102%, average 91%; EDTA plasma (n=5) 81-100%, average 90%; heparin plasma (n=5) 80-89%, average 84%.
