Sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of mouse Fibroblast Activation Protein alpha in serum, plasma, tissue homogenates, cell lysates or other biological fluids. The assay employs an antibody specific for Fibroblast Activation Protein alpha pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added to the wells, followed by a Biotinylated Anti-Fibroblast Activation Protein alpha Antibody and an HRP-Avidin conjugate. A TMB substrate solution yields a blue-coloured product that turns yellow upon addition of acidic stop solution; absorbance is read at 450 nm in a colorimetric microplate reader, and the concentration of Fibroblast Activation Protein alpha is calculated by reference to the standard curve. The assay range is 31.2–2000 pg/ml with a reported sensitivity of <13.5 pg/ml. Total assay time is 4h 30m.

Kit components include the pre-coated 96-well microplate, lyophilized standard (2 vials), Detection Solution A (120 μl), Detection Solution B (120 μl), Wash Buffer 30X (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml), and plate sealers, with components stored at +4°C or -20°C as instructed. The kit should not be used past its expiration date. Reported recovery values include serum 80–102% (average 91%, n=5), EDTA plasma 81–100% (average 90%, n=5), and heparin plasma 80–89% (average 84%, n=5). Linearity data for serial dilutions (1:2 to 1:16) are reported for serum, EDTA plasma, and heparin plasma (n=5 each).
