Mouse Farnesoid X Receptor ELISA Kit

From Antibodies.com, this Sandwich ELISA is designed for the in vitro quantitative determination of mouse Farnesoid X Receptor (Bile Acid Receptor NR1H4) in tissue homogenates or other biological fluids. The assay uses the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips. Standards and samples are captured by the immobilised antibody and detected sequentially with a Biotinylated Anti-Bile Acid Receptor NR1H4 Antibody and an HRP-Avidin conjugate. TMB substrate is then added, producing a blue product that turns yellow on addition of acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader and the target concentration is calculated from the standard curve.

The quantitative sandwich format has a reported sensitivity of 0.051 ng/ml and a working range of 0.156–10 ng/ml, with a total assay time of 4 hours 30 minutes. Matrix recovery for undiluted samples was characterised as serum (n=5) 80%–102% (average 91%), EDTA plasma (n=5) 81%–100% (average 90%) and heparin plasma (n=5) 80%–89% (average 84%); dilutional linearity was also assessed at 1:2, 1:4, 1:8 and 1:16 across the same sample types. Kit components are stored at +4°C or –20°C as instructed and the kit should not be used past its expiration date.
