Mouse Family With Sequence Similarity 135, Member B ELISA Kit

This Antibodies.com kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) for the in vitro quantitative determination of mouse Family With Sequence Similarity 135, Member B (FAM135B) in tissue homogenates or other biological fluids. The assay uses the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate arranged as twelve 8-well strips. An antibody specific for FAM135B is pre-coated onto the plate; standards and test samples are added, the wells are washed, and a Biotinylated Anti-FAM135B Antibody is added. Following incubation, an HRP-Avidin conjugate is added, then TMB substrate produces a blue-coloured product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader; the yellow intensity is proportional to the FAM135B concentration, calculated against the standard curve.

Performance: sensitivity <0.057 ng/ml, quantitative range 0.156–10 ng/ml, total assay time 4 h 30 min, colorimetric detection on a microplate reader. Recovery from spiked samples (n=5) averaged 91% for serum (range 83–100%), 95% for EDTA plasma (range 90–100%) and 96% for heparin plasma (range 91–101%). Precision at sample dilutions 1:2, 1:4, 1:8 and 1:16 was reported as: serum 90–101%, 80–108%, 84–95% and 92–104%; EDTA plasma 82–105%, 89–91%, 87–109% and 74–104%; heparin plasma 90–94%, 87–108%, 86–101% and 90–101% (all n=5). Kit components are stored at +4 °C or −20 °C as instructed and the kit must not be used past its expiration date.
