Designed for the in vitro quantitative determination of mouse PERK in tissue homogenates and other biological fluids, this sandwich ELISA from Antibodies.com employs the sandwich enzyme immunoassay technique. An antibody specific for PERK is pre-coated onto a 96-well microtiter plate, and standards or test samples are added so that PERK present in each sample is captured by the immobilized antibody. After incubation and washing, a Biotinylated Anti-PERK Antibody is added, followed by an HRP-Avidin conjugate and TMB substrate solution. The resulting enzymatic reaction produces a blue colour that changes to yellow upon addition of acidic stop solution, with the density of yellow proportional to the amount of PERK captured in each well.

The reported assay range is 0.312-20 ng/ml, with a sensitivity of <0.108 ng/ml, and absorbance is read at 450 nm in a colorimetric microplate reader. The total assay time is approximately 4 hours 30 minutes. Recovery has been evaluated in serum (n=5, average 91% across an 80% – 102% range), EDTA plasma (n=5, average 91%, 81% – 101%), and heparin plasma (n=5, average 84%, 80% – 89%). Dilution linearity has been tested at 1:2, 1:4, 1:8 and 1:16 in serum, EDTA plasma and heparin plasma.
