Mouse Elastase 2, Neutrophil ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of mouse Neutrophil Elastase in serum, plasma or other biological fluids. The kit employs a pre-coated microplate with an antibody specific for Neutrophil Elastase. Standards and test samples are added to the wells, where the target protein is captured by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-Neutrophil Elastase Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of an acidic stop solution. Absorbance is read at 450nm in a microplate reader and the concentration of Neutrophil Elastase is calculated against a standard curve.

The reported sensitivity is <0.063 ng/ml with an assay range of 0.156–10 ng/ml. The assay can be completed in 4h 30m on a colorimetric microplate reader. Validation data report average recoveries of 91% in serum (range 80%–102%, n=5), 90% in EDTA plasma (range 81%–100%, n=5) and 84% in Heparin plasma (range 80%–89%, n=5). Dilution linearity was demonstrated at 1:2, 1:4, 1:8 and 1:16 dilutions across serum, EDTA plasma and Heparin plasma samples. Kit components should be stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
