Mouse Elastase 1, Pancreatic ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit is designed for the in vitro quantitative determination of mouse Elastase 1, Pancreatic, in serum, plasma, tissue homogenates or other biological fluids.

The microplate is pre-coated with an antibody specific for Elastase 1. Standards and samples are added to the wells; following incubation and washing, a Biotinylated Anti-Elastase 1 Antibody and an HRP-Avidin conjugate are applied. TMB substrate is then added, producing a blue product that turns yellow after the acidic stop solution is dispensed. The intensity of the yellow colour is proportional to the amount of Elastase 1 captured, and the concentration is calculated from the O.D. absorbance read at 450 nm against a standard curve. The assay is colourimetric, run on a microplate reader using a pre-coated plate arranged as 12 x 8 well strips, with a total assay time of 4h 30m. Reported sensitivity is <26.5 pg/ml and the working range is 62.5-4000 pg/ml.

Spike-and-recovery performance was assessed in serum (n=5, range 80-102%, average 91%), EDTA plasma (n=5, range 81-100%, average 90%) and heparin plasma (n=5, range 80-89%, average 84%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions was also evaluated for each sample matrix. Components should be stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date.
