The assay has a product range of 0.312–20 ng/ml and a sensitivity of 0.125 ng/ml, read at 450 nm. It uses a pre-coated 96-well microplate in a 12 x 8 well strip format with a total assay time of 4 h 30 m. An antibody specific for DKK1 is pre-coated onto the plate; standards and samples are added, followed by a Biotinylated Anti-DKK1 Antibody, HRP-Avidin conjugate and TMB substrate. The intensity of the resulting yellow product is proportional to the amount of DKK1 captured, and concentration is calculated by reference to a standard curve.
