Mouse Desmocollin 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of mouse Desmocollin 1 in tissue homogenates, cell lysates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips. Standards and samples are captured by an antibody specific for Desmocollin 1 immobilised on the plate, followed by incubation with a Biotinylated Anti-Desmocollin 1 Antibody and an HRP-Avidin conjugate. TMB substrate produces a blue-coloured product that changes to yellow upon addition of acidic stop solution; absorbance is read at 450 nm in a microplate reader and the concentration of Desmocollin 1 is calculated by reference to the standard curve.

The reported sensitivity is 0.051 ng/ml with an assay range of 0.156–10 ng/ml. Total assay time is approximately 4h 30m and detection is colourimetric. Manufacturer-supplied recovery values report averages of 91% for serum (range 80–102%, n=5), 90% for EDTA plasma (range 81–100%, n=5) and 84% for heparin plasma (range 80–89%, n=5). Linearity has been demonstrated across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma.

Kit components comprise a pre-coated 96-well microplate (12 x 8 well strips), lyophilised standard (2 vials), Detection Solution A (120 μl), Detection Solution B (120 μl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and 5 adhesive plate sealers. Components should be stored at +4°C or -20°C as instructed and the kit must not be used past its expiration date. The product is intended for research use only.
