The assay uses a pre-coated microplate and employs the sandwich enzyme immunoassay technique with a biotinylated anti-ZIP Kinase detection antibody, HRP-avidin conjugate, and TMB substrate. The enzymatic reaction produces a blue colour that changes to yellow upon addition of acidic stop solution, with absorbance read at 450 nm in a colorimetric microplate reader.
