Mouse Cytochrome C Oxidase Subunit III ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com designed for the in vitro quantitative determination of mouse MTCO3 (Cytochrome C Oxidase Subunit III) in tissue homogenates or other biological fluids. The kit uses a pre-coated 96-well microplate configured as 12 x 8 well strips, with a total assay time of approximately 4 hours 30 minutes. Detection is colourimetric: an HRP-Avidin conjugate catalyses a TMB substrate to produce a blue product that turns yellow upon addition of the acidic stop solution, and absorbance is read at 450 nm on a microplate reader, with the density of yellow proportional to the MTCO3 concentration in each well.

The quantitative range of the assay is 0.312–20 ng/ml, with a sensitivity of <0.119 ng/ml. Spike-and-recovery has been validated across n=5 samples of serum (average 91%, range 80–102%), EDTA plasma (average 90%, range 81–100%) and heparin plasma (average 84%, range 80–89%). Linearity on dilution has also been characterised at 1:2, 1:4, 1:8 and 1:16 for each of the three sample types.

The capture antibody specific for MTCO3 is pre-coated on the plate, and MTCO3 in standards or samples is detected using a biotinylated anti-MTCO3 antibody. The supplied components are a pre-coated 96-well microplate, lyophilised standard (2 vials), Detection Solution A (120 μl), Detection Solution B (120 μl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml) and plate sealers (5 adhesive strips). Components should be stored at +4°C or -20°C as indicated, and the kit must not be used past its expiration date.
