Mouse Cyclophilin B ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com, designed for the in vitro quantitative determination of mouse Cyclophilin B in serum, plasma or other biological fluids. The assay employs the sandwich enzyme immunoassay technique using a 96-well microtiter plate pre-coated with an antibody specific for Cyclophilin B.

Standards and samples are added to the wells, where Cyclophilin B is captured by the immobilised antibody. After washing, a Biotinylated Anti-Cyclophilin B Antibody binds the captured analyte, followed by an HRP-Avidin conjugate. TMB substrate is then added and catalysis yields a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Cyclophilin B captured in each well; absorbance is read at 450 nm in a microplate reader and the concentration is calculated by reference to a standard curve.

The assay range is 125-8000 pg/ml with a sensitivity of <52 pg/ml, and total assay time is 4h 30m. Reported recovery in serum (n=5) is 80%-102% (average 91%), in EDTA plasma (n=5) is 81%-100% (average 90%) and in heparin plasma (n=5) is 80%-89% (average 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 is reported as 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma. The kit is supplied as a pre-coated microplate (12 x 8 well strips) with lyophilised standard, Detection Solution A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers; components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
