From Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) is designed for the in vitro quantitative determination of mouse Cross Linked N-Telopeptide Of Type I Collagen in serum, plasma, urine or other biological fluids.

The assay uses a competitive enzyme immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips. During incubation, analyte present in samples or standards competes with a fixed amount of immobilised antigen for binding sites on a biotinylated anti-analyte antibody. Unbound antibody is removed by washing, and an HRP–avidin conjugate is added to the wells. Following further incubation and washing, TMB substrate is used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow on addition of acidic stop solution. The density of yellow is inversely proportional to the amount of analyte present. Absorbance is read at 450 nm in a colourmetric microplate reader, and the concentration is calculated by reference to a standard curve.

Sensitivity is reported as 0.96 ng/ml with a product range of 3.12–200 ng/ml, and the total assay time is 2 h 30 min. Kit components should be stored at +4 °C or –20 °C as instructed and not used past the kit expiration date.
