A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of mouse Cross Linked C-Telopeptide Of Type II Collagen in serum, plasma or other biological fluids. The assay uses the sandwich enzyme immunoassay technique, with an antibody specific to the target pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, the target is captured by the immobilized antibody, and a Biotinylated Anti-target Antibody is then applied. An HRP-Avidin conjugate and TMB substrate are used to visualize the reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The intensity of the yellow colour is proportional to the amount of target captured, and absorbance is read at 450 nm in a microplate reader with concentration calculated against the standard curve.

According to the manufacturer, the product range is 15.6-1000 pg/ml and the sensitivity is <4.26 pg/ml. Total assay time is 4h 30m, and the assay is read on a colorimetric microplate reader. Recovery data are reported for serum (average 91%, range 80%-102%), EDTA plasma (average 90%, range 81%-100%), and heparin plasma (average 84%, range 80%-89%), with linearity data provided across serial dilutions from 1:2 to 1:16. Kit components include a pre-coated 96-well microplate in a 12 x 8 well strip format, lyophilized standard, Detection Solutions A and B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
