Mouse Complement Component 3 ELISA Kit

Designed for the in vitro quantitative determination of mouse Complement C3, this Antibodies.com sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is suitable for serum, plasma and other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips), where Complement C3 present in standards and samples is captured by an immobilized antibody. Following incubation and washing, Biotinylated Anti-Complement C3 Antibody is added, followed by HRP-Avidin conjugate and TMB substrate, which produces a blue product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the concentration of Complement C3 is calculated against a standard curve.

The kit has a sensitivity of <0.56 ng/ml and an assay range of 1.56–100 ng/ml, with a total assay time of 4 hours 30 minutes. In recovery testing (n=5), values were 80%–102% in serum (average 91%), 81%–99% in EDTA plasma (average 90%), and 80%–89% in heparin plasma (average 84%).

Linearity-of-dilution testing (n=5) showed the following ranges: serum 87–91% at 1:2, 87–107% at 1:4, 74–101% at 1:8, and 92–97% at 1:16; EDTA plasma 90–105%, 84–101%, 90–101%, and 79–108% at the same dilutions; heparin plasma 84–95%, 92–105%, 82–105%, and 89–91%. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
