This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of mouse granulocyte colony-stimulating factor (G-CSF, Colony Stimulating Factor 3, Granulocyte) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.

An antibody specific for G-CSF has been pre-coated onto a 96-well microplate supplied as 12 x 8 well strips. Standards and samples are added to the wells, and bound G-CSF is detected with a Biotinylated Anti-G-CSF Antibody followed by an HRP-Avidin conjugate. TMB substrate is used to visualise the enzymatic reaction, producing a yellow product whose absorbance is read at 450 nm and is proportional to the amount of G-CSF captured. The standard curve covers a working range of 15.6–1000 pg/ml with a sensitivity of <5.1 pg/ml, and the complete protocol takes 4 hours 30 minutes on a colorimetric microplate reader.

Spike-and-recovery testing (n=5) gave averages of 91% in serum, 90% in EDTA plasma and 84% in heparin plasma, with individual ranges of 80–102%, 81–100% and 80–89% respectively. Dilution linearity tested at 1:2, 1:4, 1:8 and 1:16 produced ranges of 87–91% to 92–97% in serum, 84–105% to 79–108% in EDTA plasma and 84–95% to 89–91% in heparin plasma. The kit includes a pre-coated 96-well plate, lyophilized standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and adhesive plate sealers, with components stored at +4°C or −20°C as instructed.
