Mouse Collagen Type XII ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of mouse COL12A1 in serum, plasma or other biological fluids. The assay uses a pre-coated 96-well microplate (12 x 8 well strips) immobilised with antibody specific for COL12A1. Standards and test samples are added to the wells and any COL12A1 present is captured by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-COL12A1 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The intensity of yellow is proportional to the amount of COL12A1 captured, and concentration is calculated by reading the O.D. absorbance at 450 nm in a colorimetric microplate reader against the standard curve.

The assay range is 0.312-20 ng/ml with a sensitivity of <0.106 ng/ml, and the total assay time is 4h 30m. Validation data show recovery in serum (83-100%, average 91%), EDTA plasma (90-103%, average 96%) and heparin plasma (89-102%, average 95%), with dilution linearity demonstrated across 1:2 to 1:16 in the same sample types.
