Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of mouse Factor VII in plasma or other biological fluids. The assay is performed on a pre-coated 96-well microplate with colorimetric detection read at 450 nm using a microplate reader, and the application is specifically validated for mouse reactivity.

The assay employs the sandwich enzyme immunoassay technique. An antibody specific for Factor VII is pre-coated onto the microplate. Standards and test samples are added to the wells and Factor VII present in each sample is bound by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-Factor VII Antibody is added, which binds the captured Factor VII. An HRP-Avidin conjugate is then added to the wells. After further incubation and washing, TMB substrate solution is used to visualise the HRP enzymatic reaction, producing a blue colour that changes to yellow upon addition of an acidic stop solution. The intensity of yellow is proportional to the amount of Factor VII captured in each well, and the concentration is calculated by reading the absorbance at 450 nm against a standard curve. Total assay time is 4 hours 30 minutes.

The assay range is 78.1-5000 pg/ml with a sensitivity of less than 37 pg/ml. Spike-and-recovery performance was evaluated in serum (range 80%-102%, average 91%, n=5), EDTA plasma (81%-99%, average 90%, n=5) and heparin plasma (80%-89%, average 84%, n=5). Dilution linearity from 1:2 to 1:16 was also demonstrated across serum, EDTA plasma and heparin plasma samples (n=5 each). Kit components should be stored at +4°C or -20°C as instructed.
