Description

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of mouse Uteroglobin, the protein also known as Clara Cell Protein 16, in serum, plasma, tissue homogenates, cell lysates, lung lavage fluid, cell culture supernates, and other biological fluids. The microplate is pre-coated with an antibody specific for Uteroglobin, and detection relies on a biotinylated anti-Uteroglobin antibody in combination with an HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate, producing a blue product that converts to yellow after addition of acidic stop solution; absorbance is read at 450 nm on a colorimetric microplate reader and the Uteroglobin concentration is calculated from the standard curve.

The quantitative measurement range is 78.1–5000 pg/ml with a sensitivity of <33 pg/ml, and the full protocol runs in approximately 4 hours 30 minutes. Validation data report recovery in serum (range 83–100%, average 91%, n=5), EDTA plasma (range 90–103%, average 96%, n=5), and heparin plasma (range 89–102%, average 95%, n=5). Linearity testing across serial dilutions from 1:2 to 1:16 yields recoveries generally within acceptable ranges for serum, EDTA plasma, and heparin plasma (n=5 per matrix).

Kit components include a pre-coated microplate, lyophilised standard, Detection Solution A, Detection Solution B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers. Components are stored at +4°C or –20°C as instructed in the protocol manual, and the reagents should not be used past the stated expiration date.
