Mouse Cholecystokinin A Receptor ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay is designed for the in vitro quantitative determination of mouse CCK1-R (Cholecystokinin A Receptor) in tissue homogenates or other biological fluids. The assay employs a pre-coated microplate platform configured as 12 x 8 well strips. An antibody specific for CCK1-R is immobilised on the plate; standards and samples are added, followed by a biotinylated anti-CCK1-R antibody and an HRP-avidin conjugate. The HRP enzymatic reaction is visualised with TMB substrate, producing a blue-coloured product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and concentration is calculated against a standard curve.

The assay has a working range of 0.156–10 ng/ml and a sensitivity of 0.064 ng/ml, with a total assay time of 4h 30m. Spiking recovery has been characterised in serum (n=5, range 80%–102%, average 91%), EDTA plasma (n=5, range 81%–100%, average 90%) and heparin plasma (n=5, range 80%–89%, average 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 is reported for serum (87–91%, 87–107%, 74–101%, 92–97%), EDTA plasma (90–105%, 84–101%, 90–101%, 79–108%) and heparin plasma (84–95%, 92–105%, 82–105%, 89–91%), all at n=5. Kit components include a pre-coated 96 well microplate, lyophilised standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, with storage conditions ranging from +4°C to -20°C.
