Description

A sandwich-format ELISA developed by Antibodies.com for the in vitro quantitative determination of mouse CCR7 (Chemokine C-C-Motif Receptor 7) in tissue homogenates, cell lysates and other biological fluids. The assay uses a pre-coated microplate format with read-out at 450 nm on a colorimetric microplate reader.

The kit employs the sandwich enzyme immunoassay technique. An antibody specific for CCR7 is pre-coated onto the plate; standards and test samples are added and the CCR7 present in each sample is bound by the immobilised antibody. Following incubation and washing, a biotinylated anti-CCR7 antibody is added, followed by an HRP–avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue-coloured product that turns yellow on addition of acidic stop solution. The density of yellow is proportional to the amount of CCR7 captured in each well, with the concentration calculated from a standard curve read at 450 nm.

Reported sensitivity is 0.054 ng/ml and the assay range is 0.156–10 ng/ml, with a total assay time of approximately 4 h 30 min. In spike-and-recovery testing (n=5) the average was 91% for serum (range 80–102%), 90% for EDTA plasma (81–100%) and 84% for heparin plasma (80–89%). Dilution linearity (1:2, 1:4, 1:8, 1:16) showed recoveries of 87–91%, 87–107%, 74–101% and 92–97% for serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma (n=5); and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma (n=5). Kit components are stored at +4°C or −20°C as instructed, and the product should not be used past its expiration date.
