From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of mouse Cathepsin Z in serum, plasma and other biological fluids. The assay uses a pre-coated 96-well microtiter plate arranged as 12 x 8 well strips, where an antibody specific for Cathepsin Z immobilises the target from added standards and samples. After incubation and washing, a Biotinylated Anti-Cathepsin Z Antibody is added, then an HRP-Avidin conjugate. A TMB substrate solution is used to visualise the reaction, producing a blue product that turns yellow upon addition of acidic stop solution; the absorbance is read at 450 nm in a colorimetric microplate reader and the Cathepsin Z concentration is calculated against the standard curve.

The reported assay range is 15.6–1000 pg/ml with a sensitivity of 7.1 pg/ml, and the total assay time is 4h 30m. Sample recovery (n=5) is reported at 80–102% (average 91%) in serum, 81–100% (average 90%) in EDTA plasma and 80–89% (average 84%) in heparin plasma. Components are stored at +4°C or -20°C as instructed, and the product is for research use only; it is not intended for diagnostic or therapeutic use.

The protocol also supports sample preparation from cell culture supernatants, tissue homogenates, cell lysates and other biological fluids, with guidance on collection, centrifugation and storage provided. Required but not supplied materials include a microplate reader capable of measuring absorbance at 450 nm ± 10 nm, a multichannel pipette reservoir or automated microplate washer, computer software capable of generating logarithmic functions, test tubes and Eppendorf tubes, multi- and single-channel pipettes with sterile tips, deionized or distilled water, absorbent paper towels and a 37°C incubator.
