A sandwich ELISA kit from Antibodies.com designed for the in vitro quantitative determination of mouse Cathepsin B in serum, plasma, tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique using an antibody specific for Cathepsin B that is pre-coated onto a microplate. Standards and test samples are added to the wells, where Cathepsin B is bound by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-Cathepsin B Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Cathepsin B captured in each well, with absorbance read at 450 nm in a colorimetric microplate reader and concentration calculated from a standard curve.

The kit has a sensitivity of <0.057 ng/ml and an assay range of 0.156-10 ng/ml, with a total assay time of 4h 30m. Spike-and-recovery testing (n=5) gave recoveries of 80-102% in serum, 81-100% in EDTA plasma and 80-89% in heparin plasma. Sample dilution linearity was demonstrated at 1:2, 1:4, 1:8 and 1:16 dilutions across the same sample types. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
