Mouse Caspase 8 ELISA Kit

A sandwich enzyme-linked immunosorbent assay (sELISA) from Antibodies.com, this Mouse Caspase-8 kit enables the in vitro quantitative determination of mouse Caspase-8 in tissue homogenates, cell lysates, cell culture supernates, and other biological fluids. The assay uses a 96-well microtiter plate pre-coated with a Caspase-8-specific capture antibody; standards and test samples are added and Caspase-8 is bound by the immobilised antibody. Following incubation and washing, a biotinylated anti-Caspase-8 detection antibody is added, followed by an HRP-avidin conjugate and TMB substrate. The HRP-catalysed reaction produces a blue product that turns yellow upon addition of acidic stop solution, with absorbance read at 450 nm in a microplate reader.

The standard curve spans 0.156–10 ng/ml, with a sensitivity of 0.069 ng/ml. Total assay time is 4 hours 30 minutes on a pre-coated 12 × 8 well strip microplate with colourimetric detection. Spike-and-recovery ranges (n=5) are 80%–102% (average 91%) for serum, 81%–100% (average 90%) for EDTA plasma, and 80%–89% (average 84%) for heparin plasma. Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 in serum, EDTA plasma and heparin plasma. Components are stored at +4°C or –20°C as instructed and the kit should not be used past its expiration date.
