Mouse Calpain 1, Large Subunit ELISA Kit

From Antibodies.com, this sandwich ELISA kit enables the in vitro quantitative determination of mouse Calpain 1 in tissue homogenates, cell lysates, cell culture supernates, or other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for Calpain 1, with standards and test samples added to the wells and bound by the immobilised antibody. Detection is performed using a Biotinylated Anti-Calpain 1 Antibody and an HRP-Avidin conjugate, with a TMB substrate producing a colour change that is read at 450 nm in a colorimetric microplate reader.

The kit reports a product range of 1.56–100 ng/ml and a sensitivity of 0.58 ng/ml. Total assay time is 4 h 30 min. Spike-and-recovery across sample types has been observed in the following ranges: serum 80–102% (average 91%), EDTA plasma 81–100% (average 90%), and heparin plasma 80–89% (average 84%). Dilution linearity at 1:2, 1:4, 1:8, and 1:16 dilutions has been demonstrated for serum (74–107%), EDTA plasma (79–108%), and heparin plasma (82–105%).

Kit components include a pre-coated 96-well microplate (12 x 8 well strips), lyophilised standard, Detection Solutions A and B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers. Storage temperatures differ by component, with instructions to keep reagents at +4°C or −20°C as indicated on the package leaflet and to avoid use past the expiration date.
