From Antibodies.com, this Sandwich ELISA (sELISA) kit is designed for the in vitro quantitative determination of mouse Calcium Sensing Receptor (CaSR) in tissue homogenates, cell lysates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for CaSR pre-coated onto a 96-well microtiter plate. Standards and samples are added to the wells where CaSR is bound by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-CaSR Antibody is added, followed by HRP-Avidin conjugate. TMB substrate solution is used to visualize the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of CaSR captured, and concentration is calculated by reading the O.D. absorbance at 450nm against a standard curve.

The reported sensitivity is <0.057 ng/ml and the assay range is 0.156–10 ng/ml. Reported sample recovery values include serum at 80–102% (average 91%), EDTA plasma at 81–100% (average 90%), and heparin plasma at 80–89% (average 84%). Linearity performance at 1:2, 1:4, 1:8 and 1:16 sample dilutions is reported for serum, EDTA plasma and heparin plasma. The assay is read on a colorimetric microplate reader, takes 4h 30m, and is intended for research use only. Kit components include a pre-coated 96-well microplate, lyophilised standard, Detection Solutions A and B, Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers, with storage at +4°C or -20°C as instructed.
