A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of mouse VE Cadherin in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique, with an antibody specific for VE Cadherin pre-coated onto a 96-well microtiter plate. Standards and test samples are added into the wells, after which the wells are washed and incubated with Biotinylated Anti-VE Cadherin Antibody. Following incubation, HRP-Avidin conjugate is added, and after further washing, TMB substrate solution produces a blue colour that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of VE Cadherin captured, and the concentration is calculated by reading O.D. absorbance at 450 nm against a standard curve.

The product range is 0.156-10 ng/ml with a sensitivity of 0.051 ng/ml. The assay is quantitative and colorimetric, run on a pre-coated microplate platform of 12 x 8 well strips, with a total assay time of 4h 30m. Results are read using a colorimetric microplate reader. Kit components are stored at +4°C or -20°C as instructed and should not be used past the expiration date.

Validated recovery (n=5) averages 91% for serum (range 80%-102%), 90% for EDTA plasma (range 81%-100%) and 84% for heparin plasma (range 80%-89%). Linearity across dilutions of 1:2, 1:4, 1:8 and 1:16 has been demonstrated: serum ranges 87-91%, 87-107%, 74-101% and 92-97%; EDTA plasma ranges 90-105%, 84-101%, 90-101% and 79-108%; heparin plasma ranges 84-95%, 92-105%, 82-105% and 89-91%.
