Mouse CA199 ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of mouse CA199 in serum, plasma, tissue homogenates, and other biological fluids. The assay uses a pre-coated microplate with an immobilised capture antibody specific for CA199. Standards and samples are added to the wells, followed by a biotinylated detection antibody that forms an antibody–antigen–antibody sandwich complex. After incubation and washing steps, an HRP-Streptavidin conjugate and TMB substrate are added; the resulting blue product turns yellow upon addition of the acidic stop solution. Absorbance is read at 450 nm, with the yellow intensity directly proportional to the CA199 concentration, which is calculated against a standard curve.

The manufacturer specifies an assay range of 0.625–40 IU/mL and a sensitivity of <0.375 IU/mL, with no reported cross-reactivity with other analogs or variants. Average recovery across 10 samples per matrix is reported as 94% for serum, 94% for EDTA plasma, and 100% for heparin plasma, with corresponding ranges of 85–103%, 86–105%, and 90–104%. Dilution linearity at 1:2, 1:4, and 1:8 across 10 samples per matrix is listed as 91–105%, 89–101%, and 86–102% for serum; 83–96%, 86–99%, and 86–96% for EDTA plasma; and 84–99%, 82–99%, and 80–97% for heparin plasma. The complete assay time is approximately 4 hours, and the kit is intended for use on a colorimetric microplate reader.

The kit is shipped at +4°C, and components are stored at +4°C or –20°C as instructed in the protocol manual. The manufacturer advises avoiding freeze/thaw cycles and not using the kit past its expiration date.
