From Antibodies.com, this competitive Enzyme-Linked Immunosorbent Assay (cELISA) is designed for the in vitro quantitative determination of mouse C-Peptide in serum, plasma, tissue homogenates, cell lysates, cell culture supernates, or other biological fluids.

The assay employs the competitive enzyme immunoassay technique on a 96-well microplate pre-coated with C-Peptide antigen. During incubation, C-Peptide present in samples or standards competes with the fixed amount of immobilized C-Peptide for binding sites on a biotinylated anti-C-Peptide antibody. Following washing to remove unbound conjugate, an HRP-avidin reagent is added and TMB substrate produces a colour change inversely proportional to C-Peptide concentration. The standard product range is 78.1-5000 pg/ml with a sensitivity of <19.5 pg/ml. Absorbance is read at 450 nm in a microplate reader. Total assay time is approximately 2 hours 30 minutes, and the microplate platform comprises 12 x 8 well strips.

Recovery has been evaluated across serum (average 91%, n=5), EDTA plasma (average 90%, n=5), and heparin plasma (average 93%, n=5), with dilution linearity ranges from 1:2 to 1:16 also reported. Kit components include the pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers, with storage between +4°C and -20°C as instructed.
