Mouse BH3 Interacting Domain Death Agonist ELISA Kit

Antibodies.com offers this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) intended for the in vitro quantitative determination of mouse Bid (BH3 Interacting Domain Death Agonist) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The kit is reactive with mouse samples and uses pre-coated capture antibody and biotinylated detection antibody chemistry.

Quantitative performance is defined by a product range of 0.312-20 ng/ml and a sensitivity of 0.112 ng/ml. Detection is colourimetric, read as optical density at 450 nm on a microplate reader following catalysis of TMB substrate. The total assay time is 4 hours 30 minutes. The principle involves a Bid-specific antibody immobilised on the microplate, incubation with standards and samples, binding of biotinylated anti-Bid antibody, addition of HRP-Avidin conjugate, and visualisation with TMB substrate that produces a blue product changing to yellow upon addition of acidic stop solution; the absorbance at 450 nm is proportional to captured Bid and is quantified against a standard curve.

The assay is validated for serum, plasma, tissue homogenates, cell lysates and cell culture supernates. Recovery testing in serum (n=5) gave an average of 93% across a range of 86-100%, in EDTA plasma (n=5) an average of 94% across 87-101%, and in heparin plasma (n=5) an average of 94% across 88-100%. Kit components are stored at +4°C or -20°C as instructed.
